Tonsillar B cells at a concentration of 1 × 106/mL in supplemented RPMI1640 were treated with 400 U/mL recombinant IL-4 plus 1 μg/mL anti-CD40 in the presence or absence of omalizumab or control antibody (human IgGκ). Omalizumab was reconstituted in sterile PBS at a concentration of 15 mg/mL. Dilutions of omalizumab were made in RPMI1640. In some instances the cells were pre-treated with FcR blocking reagent. At the indicated times, viable cell numbers were enumerated by their ability to exclude the dye trypan blue. Culture supernatant fluid was harvested for IL-6 quantification by ELISA.
